Queen Elizabeth Way is closed through Burlington between Appleby Line and Burloak Drive in both directions after a collision.
OPP said at least one man is reported dead.
Vehicles are being diverted onto municipal roads in both directions around the investigation site.
Drivers can expect long delays through the area and are being asked to use alternate routes if possible, Halton police said.
Police aren't reporting any other details at this time.
